Amplification of a Recombinant Adenovirus
Description
Generation of a high-titer viral stock from a recombinant adenovirus.
Plaque purification prior to amplification is highly recommened
to insure the purity and stability of the recombinant adenovirus.

Time Required: 4-6 weeks
Includes:
- Amplification of a recombinant adenovirus in QBI-HEK 293A cells
at the desired volume.
- Collection of the adenovirus. Infected cells are harvested
and concentrated. Three freeze/thaw cycles are performed and cell
debris is removed by centrifugation.
- Delivery of supernatant containing recombinant adenovirus.
The final titer is variable and depends on the nature of the recombinant
adenovirus to be amplified. Typically, QBI-HEK 293A cells will produce
between 1,000 and 10,000 viral particles per cell. For an accurate
infectious titer, a TCID50 (ACS0070) or plaque assay
(ACS0060) can be performed on the final product.
