Telomerase Probes (hTERT;hTERC)

A set of probes for 3q26 (hTERC) and 5p15 (hTERT) is now available from Qbiogene, manufactured using the Universal Linkage System (ULS )

These direct labeled probes have been tested and qualified on cultured cells, blood smears and paraffin-embedded tissue
sections.

3q26 hTERC - Human RNA component for Telomerase
A common amplification target of encompassing chromosome region 3q25 to q27 has been identified by comparative genomic hybridization analyses in tumors of the cervix, ovary, endometrium, lung, and head and neck. The region of highest amplification included the gene for the RNA component of telomerase (hTERC), supporting it as a potential target. Although the importance of low-level amplification is unknown, the consistent and reproducible nature of this event in a variety of carcinomas suggests that 3q26.2 harbors an oncogene whose low-level amplification has a significant influence on tumor biology. (Cancer Genet Cytogenet 2000 Feb;117(1):9-18)

5p15 hTERT – Catalytic subunit of Human Telomerase
The gene for the reverse transcriptase component of telomerase (hTERT) has been mapped by fluorescence in situ hybridization (FISH) to chromosome 5p15.33. Increase in copy number of the hTERT gene region do occur, and may well be one route to upregulating telomerase levels in tumour cells. 5p15 gains and amplifications have been documented for various tumour types, including non-small cell lung carcinoma, squamous cell carcinoma of head and neck, and uterine cervix cancer, making hTERT a potential target. (Oncogene 1998 Mar 12;16(10):1345-50)
The 5p15 hTERT probe is also available in combination with a 5q31(D5S89) specific probe as dual-color assay. The 5q31 probe can serve as internal control to verify deletions or amplification of 5p15.
