
Getting
Started: Reporter AdenoExpress
Prior to
starting a recombinant adenovirus production we recommend you test your
specific in vitro or in vivo model with either GFP or LacZ Reporter AdenoExpress.
These are ready-made, high titer recombinant adenoviruses that serve to
optimize transduction parameters such as determining the optimal MOI (Multiplicity
of Infection) needed to infect a specific cell type as well as the amount
of virus that will be required to perform the entire research project
(the level of large-scale production).
Reporter
AdenoExpress can also be used as a control in experimental
protocols. GFP offers an advantage over LacZ in that it can be used in
real time assays in living cells, whereas LacZ can be detected by a simple
colorimetric assay.
Transduction
efficiencies vary with the number of Adenovirus cell surface receptors
(the CAR, or "Coxsackie virus and Adenovirus Receptor", and the vß3 or vß5 integrins), which
differs greatly in number from one cell line to another. Preliminary experiments
and evaluation studies using AdenoExpress represent the most
reliable and convenient methods of getting started with adenoviral vector
technologies.

Reporter AdenoExpress two titers are available:
- High Titer
(1x1011 VP/mL) for gene transfer in vitro
- Very High
Titer (1x1013 VP/mL) for gene transfer in vivo
