Capture mRNA with the high-affinity Poly T gripNAT Probe instead of using oligo dT.

PNAs are DNA analogs in which the four nucleotides are attached to a backbone different from the familiar deoxyribose phosphate backbone found in DNA. This remarkable chemistry allows PNAs to hybridize to complementary DNA and RNA sequences with significantly higher affinities than those of corresponding DNA oligonucleotides (oligo dT). The Poly T gripNA Probe found in the mTRAP™ mRNA Isolation Kits is a patent-pending hybrid PNA molecule offering distinct advantages over oligo dT-based purification of mRNA.

Poly T gripNA Probe has a Higher Binding Affinity than Oligo dT

  • mRNA isolation can be performed in buffers with lower salt concentrations.
    • Eliminate secondary structure that can make mRNA inaccessible for capture.
    • Eliminate non-specific bonds that can cause contamination with rRNA and protein.
  • mRNA is selectively eluted away from unwanted genomic DNA
  • mRNA with short (< 20 bases) poly A tails are more available for capture

Poly T gripNA Probe Not Susceptible to Enzymatic Digestion like Oligo dT

  • Optional DNase step can be performed if absolute control of genomic DNA is required for downstream applications

 

   Basic mRNA Isolation Procedure:

  1. Homogenize tissue or cells in Lysis Solution
  2. Add Poly T gripNA Probe and incubate
  3. Add Streptavidin Beads, incubate and wash
  4. Elute mRNA from beads and precipitate

mTRAP™ Kits are available in two sizes (Midi and Maxi) for purification of mRNA directly from tissue or cells, and in one size for the purification of mRNA from total RNA. All kits include required plasticware and RNase-free solutions.
